Description
Kisspeptin is a synthetic peptide identical to the biologically active C-terminal fragment of the natural human KISS1 gene product. The most commonly used therapeutic form is Kisspeptin-54 (also known as metastin), a 54-amino-acid peptide with the sequence: NH₂-Tyr-Asn–Trp–Asn-Ser-Phe-Gly-Leu-Arg-Phe-NH₂ (amidated C-terminus). Shorter active fragments (e.g., Kisspeptin-10) retain full potency and are sometimes employed. The peptide contains a critical C-terminal amidated motif essential for receptor binding.
Mechanism of Action
Kisspeptin is the most potent known stimulator of gonadotropin-releasing hormone (GnRH) neurons in the hypothalamus. It binds with high affinity to the KISS1R (GPR54) G-protein-coupled receptor expressed on GnRH neurons, triggering:
- Rapid depolarization and increased firing rate of GnRH neurons
- Pulsatile release of GnRH into the hypophyseal portal system
- Subsequent stimulation of pituitary gonadotrophs → secretion of luteinizing hormone (LH) and follicle-stimulating hormone (FSH)
- Downstream gonadal steroidogenesis (testosterone in males, estradiol/progesterone in females)
Kisspeptin therefore acts as the primary upstream regulator of the hypothalamic-pituitary-gonadal (HPG) axis, controlling reproductive hormone secretion, puberty onset, fertility, and reproductive endocrine homeostasis.
Indications & Usage
Hormone Regulator (Kisspeptin) is used to support physiological regulation of the reproductive endocrine axis. It is indicated for:
- Restoration or enhancement of pulsatile GnRH/LH/FSH secretion
- Support for reproductive hormone balance in age-related decline or stress-induced suppression
- Assistance in recovery of hypothalamic-pituitary-gonadal function
- General support for vitality, libido, and endocrine resilience
Contraindications & Warnings
Do not use if you have:
- Known hypersensitivity to kisspeptin or any excipient
- Hormone-dependent malignancy (e.g., prostate cancer, breast cancer, endometrial cancer)
- Severe uncontrolled endocrine disorders (e.g., hypergonadotropic hypogonadism with high LH/FSH)
- Pregnancy or breastfeeding
- Age under 18 years (unless under specialist paediatric endocrinology care)
Warnings
- Use only under medical supervision with regular monitoring (LH, FSH, testosterone/estradiol, SHBG, PSA in males, clinical symptoms).
- Not a substitute for standard medical treatment of hypogonadism, infertility, or endocrine disorders.
- Potential for overstimulation of the HPG axis (e.g., excessive LH/FSH release, ovarian hyperstimulation in females).
- Discontinue immediately and seek medical attention if severe symptoms occur (e.g., severe headache, visual disturbances, abdominal pain, or signs of allergic reaction).
Dosage & Administration
- Subcutaneous injection only.
- Standard Dose Range: 100–200 mcg per injection (common effective range)
- 100 mcg ≈ 12 clicks
- 200 mcg ≈ 24 clicks
- Frequency: Once daily or as directed by your healthcare provider
- Timing: Consistent daily timing (morning or evening preferred)
Administration Instructions:
- Wash hands and prepare a clean surface.
- Attach a sterile needle.
- Dial the required number of clicks.
- Clean injection site (abdomen or upper thigh) with alcohol swab.
- Insert needle at 45–90° angle and inject slowly.
- Withdraw needle, dispose safely, and cap the pen.
- Rotate injection sites to minimize irritation.
- Always inspect solution before use: Do not inject if cloudy, discoloured, or containing particles.
Side Effects
Kisspeptin is generally well tolerated at physiological replacement doses. Reported reactions are uncommon and usually mild/transient, including:
- Mild injection-site irritation, redness, or itching
- Transient headache or light-headedness
- Temporary flushing or warmth sensation
- Mild gastrointestinal upset (nausea, abdominal discomfort)
- Rare transient changes in libido or energy levels
If any severe or persistent symptoms occur (e.g., severe headache with visual changes, severe abdominal pain, or signs of allergic reaction), discontinue use and contact your healthcare provider immediately.
Storage & Handling
- Store either in refrigerator (fridge); never freeze or in a cool dry place (≤25–30°C / 77–86°F).
- Keep pen capped and protected from light and heat.
- After first use: Same storage conditions.
- Keep out of reach of children.
- Discard if expired or solution appears abnormal.
